Objectives

Knowledge and Understanding
Knowledge of specialistic methodologies for the rigth solution definition and study of complex problems endowed with biotechnology process engineering;
Ability to understand the issues related to health, security, and environmental impact of biotechnological processes.
Applying Knowledge and Understanding
Ability to apply advanced instruments and technologies for the development and management of biotechnological processes;
Ability to utilize the acquired knowledge for the innovation of processing industry
Ability of decide on, study and apply advances methodologies to biotechnological process control.

Prerequisites

Good knowledge of Organic Chemistry and the fundamental concepts of Biochemistry, Physiology, General Pharmacology.
Requirements:
for admission to the examination, the student must have passed the examination of "Organic Chemistry 2"

Contents

Biotechnology classification. Traditional and advances biotechnologies. History of biotechnology. Biotechnology applications. Biotechnology products.
Gene cloning. DNA fragmentation. Restriction endonuclease. Cloning vectors. Gene transfer technologies.
Transfection: chemical, physical and biological methodologies. Recombinant selection. Products of recombinant DNA technology. Basic knowledge of PCR and its applications.
Fermentation classification. Steps of industrial fermentation processes. Bioreactor configuration and control systems.
Microbial biomasses, primary metabolites, secondary metabolites, enzymes, complex microbial products, products of biotransformation reactions. Strategies for microbial metabolites production.
Media and raw materials
Microbial nutrition (macronutrients, micronutrients, growth factors and vitamins). Synthetic and complex media: C and N sources.
Alcoholic fermentation, lactic fermentation. Production of citric acid. Production of lactic acid. Production of amino acids. Production of enzymes. Production of vitamins. Production of antibiotics. Production of proteins. Single cell protein. Stability problems. Production of proteins by recombinant DNA technology. Post-traductional modifications and folding. Human insulin production from escherichia coli and saccharomices cerevisiae.
Production of amino acids and peptides by enzymatic processes. Enzyme catalyzed reactions. Lipase catalyzed reactions. Glycosidase catalyzed reactions. Enzymatic hydroxylation reactions. Enzyme manipulation. Peroxydase catalyzed oxidations. Dioxygenases. Decarboxylases.
Production methodologies, characteristics and applications of monoclonal antibodies. Murine, chimeric, humanized and human antibodies production and use.
Biopolymers.

Teaching Methods

The course consists of lectures (60 hours). Lectures will be prevalently held in classrooms, also integrated with online teaching resources, by using specific online platforms managed by the University of Cagliari

Verification of learning

With reference to course objective the Assessment is carried out by a final oral examination during which the student will demonstrate the knowledge of specialist methodologies for the right solution definition and study of complex problems endowed with biotechnology process engineering as well as in relation to related to health, security, and environmental impact. The student will demonstrate the ability to utilize the acquired knowledge for the innovation of processing industry and to apply advances methodologies to biotechnological process control. Furthermore the student will be demonstrate to have an excellent command of general and biotechonology linguistics, as well as a sufficient synthesis and critical judgment ability. The final mark is defined by means of a weighted combination of the marks in the specific topics.
